For example breasts enhancement, reshaping of the nose etc.This country is perfect for our organization because of the demand of reputable and professional plastic surgeons.   This is due to the increase of unscrupulous people who refer themselves as plastic surgeons for the sake of making money from their patients while not doing the surgery we.   There has been an increase in cases, where some consumers have undergone a cosmetic surgery.For example tummy tucks and have ended-up with more surgical complications e.g. incomplete tummy tucks due to infection.The public relations will focus in communicating the positive side of cosmetic surgery, for example most people assume that this industry is only meant for the rich, celebrities e.g. musicians, actress but this is not true.   The ageless will offer cosmetic surgery at affordable prices for its consumers compared to the other practitioners, hence anyone seeking for the service can be able to afford it.The cosmetic surgery helps people to fix any birth defects or abnormalities that the potential   consumer is uncomfortable with e.g. reshaping of the nose, face lift which will be preformed incase where there is premature formation of wrinkles due to financial stress, effects of facial creams etc.The public realtions has decided to focus their main attention of communication to the female gender.   This is because this the largest and willing consumer who opt for cosmetic surgery to fix their physical problems.   The communication strategies will mainly focus on women who have already given birth.   After careful analysis, it was found out women who have given birth seek for affordable cosmetic surgery to help them fix some of the aftermath physique problems for example decrease of stretch marks by use of raiser dermatology technique removal of excess skin especially at the abdominal area etc.Most of the market segments i.e. the women; it has been observed that immediately after giving birth, they gain baby fat of which after vigorous exercise plan they are able to shed-off the baby fat still resulting to loose skin, the breasts drooping and this shatters their self-esteem. Eliza’s Transformation in Pygmalion by George Shaw Essay

In the play Pygmalion by George Shaw, Eliza experiences a type of transformation. Before Eliza first encountered Mr. Higgins, she was a dirty, improper, poor young girl. During her time with both Mr. Higgins and Colonel Pickering, Eliza did change. Her change seems so go in somewhat of a cycle, however. For the fist few weeks of her stay she questioned everything that Higgins asked her to do. She simply was unable to see how they would help her. Later, Eliza begins to understand that even though Higgins’ ways are so harsh, he is doing his best to teach her and he deserves some cooperation. After the ambassador’s ball, we see more of the old Eliza because her task is finished. She starts to worry again, and since she has grown attached to Higgins and Pickering, she is so upset that they still see her as something of little value. Eliza’s upper-class speech and manners can be observed, but her inner confidence is what changes the most. In the beginning of Higgins’s study, Eliza feels that she has to impress Higgins by making sure he knows that she arrived in a taxi. Eliza does not understand Higgins’ personality at first and feels like he is being specifically mean to her. During her lessons, Eliza is worked so hard she begins to regret allowing herself to put up with Higgins as a teacher. Her hatred towards the man disappears a little bit when she realizes that she can only accomplish her dream of working as a lady in a flower shop if he is shapes her into a lady. She tried to hide the anger built up because of Higgins and think of him as more of a friend. She does not realized at first that Higgins takes pride in his work and not his student. She soon realized that Higgins’ investment in her was merely a tool used to enhance Higgins reputation in society. Higgins showed no appreciation towards Eliza as he repeatedly boasted about his success, and he not once acknowledged her. Higgins was able to transform Eliza into a lady, but what he did not realized was that he had slowly been making Eliza a stronger person. During the process, his constant beating on Eliza made her realize that she did not deserve his disrespect. She begins to stand up for herself and believe in her worth. This is illustrated in Eliza’s throwing the slippers at Higgins. Eliza finally stands up to Higgins and does it in a bold way. Higgins was able to change Eliza. Originally she was an innocent girl trying to stay alive. Higgins through the introduction to a proper life-style had altered Eliza’s way of thinking. It was good for Eliza to become stronger as she did. At the end of the play, she becomes overpowering to Higgins, her beauty becomes irresistible as Higgins realizes that she is leaving. It took the threat of Eliza leaving for him to see his true feelings towards her. Eliza’s strength is shown the most when she is finally able to leave Higgins. She shows that she is able to take care of herself and does not need Higgins’ superficial changes to survive.

‘Investigating Wakefield’ – GCSE Geography Coursework

The overall topic of this investigation is land use within settlements. Urban land areas in towns and cities have got very complicated land use patterns and the industrial city of Wakefield is no exception. It has grown and developed over hundreds of years and now has a thriving population of around sixty thousand. Wakefield is a city situated in the North-East of England. It has good access to the M1 and A1 which is why it is such a popular destination for many factories and businesses. Wakefield is nine miles from the major city of Leeds which is also in Wakefield's county of West Yorkshire. The hypothesis which was investigated was ‘Are there obvious land use zones along a transect from the edge of Wakefield to the Central Business District (CBD)?' and to do this a transect of land which was 1.5km long was investigated. The transect started at Newton Bar and finished at the end of Wood Street, therefore coming in to the city centre in a southerly direction. Method A transect of land use from the city centre to the Northern outskirts of Wakefield was recorded. The transect was 1.5km long, stretched from Newton Bar roundabout to the end of Wood Street and the land use immediately next to the road was recorded by hand in a notebook. The age of the buildings was estimated and if any dates were visible they were recorded too. In Wood Street, some of the buildings had more than one use and both these uses were observed and recorded. After the data had all been collected, a land-use transect was drawn. Eight different colours were used to represent the different land use categories and this helped to decipher patterns in the land use. The land use categories which were used were residential, public buildings and services, healthcare, offices, entertainment, retail, education and religion because then the buildings were easier to categorise and then record. A simplified model was also produced to show the main land use zones more clearly. Description of the Area Studied The area of Wakefield which was used to carry out the hypothesis on was a transect of 1.5km and stretched from Newton Bar Roundabout to the end of Wood Street in the CBD. Newton Bar is about one mile from the CBD and the transect studied enters the city in a southerly direction. The area at the beginning of the transect has terraced housing and is slightly run down but industrial because it has a council construction site and a development department in close proximity, and a new housing estate is now being built. The quality of buildings improves as one goes further along the transect towards the CBD, the houses on St. John's North, Wentworth Street and Wentworth Terrace are particularly smart and elegant. As one walks down Bond Street and Wood Street, one sees buildings such as the County Hall, Crown Court, Police Station, Wakefield Museum and Town Hall which are superb examples of magnificent architecture. When one reaches Cross Street, the more commercial part of Wakefield begins with lots of small shops and businesses such as cafes, hairdressers and estate agents. There are also bars and other forms of entertainment such as internet cafes as well as ‘quality' stationers, book shops and music shops. This area of Wakefield is quite clean and well maintained although some of the buildings were probably built in the 1960s and are therefore less attractive. The pictures below show some of the different land use in places along the transect. Results The results were recorded by hand in a notebook as the transect's land use was observed and then a land use transect from Newton Bar to Wood Street was drawn as I have described in my method above. A simplified model was also drawn later to show the main land use zones more clearly. This model shows that there are five main zones along the transect, each being different in size and length. The zones are, coming out in order from the CBD, retail, public buildings/services and offices, education and healthcare, residential and public buildings/services. These zones reflect the types of land use there are in Wakefield. Discussion – Interpretation and Analysis of Results Zone 1 Zone one on my simplified model of land use is retail. This zone is in the middle of the CBD, starts at Cross Street and ends at the end of Wood Street. The land use there is mostly for shops and small public services or businesses. There are cafes, hairdressers, estate agents and lots of other small shops. These types of businesses are in the CBD because it is a typical type of zone to find in the middle of the city and because of accessibility. The businesses within this zone need to be in the CBD because their customers need to be able to get to them easily and if they are in the CBD there are lots of easy ways for them to do this – for example public transport and nearby car parking. It is good for these land uses to be in the CBD because it is the most accessible place to be situated for customers and they receive a lot of deals with passers-by who happen to go into their shop/business/public service or else inform other people about it. However, as the land prices are usually high in the CBD, only successful businesses are able to afford it. In Wood Street most of the buildings are old terraced houses which could also be a reason as to why these businesses are situated there, because lots of the buildings in Wood Street had flats or offices on the second story of their building and so would want buildings with more than one story. This type of zone would also be found in other cities, as it is a typical zone of a CBD. Zone 2 Zone two of the model consists of public buildings/services and offices. This zone begins at Bell Street and ends at Chancery Lane and included in this zone there are buildings such as the County Hall, the Crown court, the Town Hall, Wakefield Museum and the Police Station and offices. These buildings are where they are because they couldn't be in the CBD as they would not have been able to afford the land prices but they still needed to be near it so that they are accessible for visitors and workers. They are also on the major road to Leeds so they have good transport links. Another reason for these buildings being there could be so that the council could keep all its buildings together and so that everyone would be able to find the council buildings easily, without having to travel all over Wakefield to find the one they wanted. The buildings which are in this zone are all old, the County Hall was built in 1898 and many of the buildings have pillars outside the building which implies that they were built quite a long time ago. However, if the people who work in these places have to pay for the upkeep of the buildings, then this could be why they are not situated in the middle of the CBD – they do not have enough money to purchase land in the city centre. This type of zone could be found elsewhere in Wakefield because only one transect was investigated, which means there would be other transects possible to investigate on the other side of the CBD which implies that there could be other places in Wakefield with similar land use patterns. Zone 3 Zone three from the model is education and healthcare and in this zone there is Wakefield College, Wakefield Girls' High School, Clayton Hospital and an 1871 dentist. This zone also includes Wakefield Art Galley and the Christian Science Building. This zone stretches form Bell Street to North Road and is in the place it is because it, again, needs to be accessible, this time for students and workers, and because the land uses in it will not have been able to afford the high land prices of the CBD. Even if this zone could have been situated in the CBD, it would not have been because hospitals and schools need space to expand their premises and if they were in the CBD there would be no space to do this. Also, hospitals and schools need to have lots of space for car parking for workers and/or students and there would be no space for this in the CBD. The hospital and school also need good transport links in order for its workers/students to get there each morning which is another reason for why they are in this zone, it is on a major road which leads to the large city of Leeds. This zone could be found in other major cities such as Leeds, but not elsewhere in Wakefield. Zone 4 Zone four begins at the Leeds Road and stretches till North Road. In zone four the land use is mainly residential, but there are also places of religion and education such as St. John's Church, built in 1791, and St. John's Primary School, built in 1964. The housing in zone four is very varied, because there are areas of terraced houses from the 1920s-30s such as Snow Hill View, the 1960s-70s as in the houses between Andrew Street and St. John's Avenue and Georgian terraced housing in St. John's Square which is a protected heritage site. These buildings are where they are because they could not be in the CBD as it is far too expensive but they still needed to be easily accessible from the city centre. St. John's Square will have been built for wealthy people to live near the Church as the Church would have been the centre of the community in those times. St. John's C of E School, which was originally on Kilby Street (named after the Reverend Thomas Kilby), was rebuilt nearby as it is a Church school. The houses from the 1960s-70s were built on waste land which would have been cheap at the time but still had good transport links with the city centre, Church and local schools. This zone could not be found elsewhere in Wakefield but a similar area might be found in other cities. Zone 5 Zone five is between the Leeds Road and Newton Bar and it consists of mainly public buildings/services, although there are some terraced houses and quite a lot of open space. Included in this zone are ‘The Green', a council construction site, the Development Department, and the Vine Tree public house. These buildings are where they are because they could definitely not have been in the CBD, as they require a lot of land and room for expansion which would not be available in the city centre. However, even if there was suitable land available for these land uses in the CBD, it would have been too expensive for the businesses to afford and so they would have been forced to build further away from the centre anyway. As many of the workers in these buildings would come to work by car, these buildings also require space for car parks and good transport links, both of which they have. This zone would probably not be found anywhere else in Wakefield but it is very likely that other cities have parallel zones. Conclusion I found that, in Wakefield, there are five main land use zones and that as one goes further away from the city, the amount of housing increases. There is also more open space further away from the CBD which is not surprising as most businesses want to be in the city centre so their customers can have easy access and so that they get trade with people who happen to pass by. I also found that there seemed to be a greater concentration of shops and offices in the CBD but that there were also quite a lot of public buildings/services as well. There were no residential areas in the CBD which suggests that people prefer to live outside the city centre and go in each morning for work. I think that Wakefield's land use zones are quite similar to the Sector Model of land use so this suggests that Wakefield is a typical city. Evaluation I did this investigation to find out if there are obvious land zones along a transect from the edge of Wakefield to the CBD and I found that there were. Overall I think that the investigation went well but that it could have been better if I had not had to go out to study the transect on two different days. If I was to do this investigation again, I would investigate the transect once, in one session, and then I would go out at another time to record the landuse again. This would make my results more accurate because if I did the investigation twice, I would be able to match up the landuse on each set of my results and make sure I had not missed anything out the first time.

The plantations were fully self contained with their own blacksmith, teachers and professionals. So there were no big cities or towns. The Southern Colonies had a strict three class system: upper class rich plantation owners, middle class small plantation owners, lower class poor whites and a population of Negroes of no class. As can be seen socially all three sets of colonies were different. Yeoman farmers, who worked smaller tracts of land, sat in popular assemblies and found their way into political office. Their outspoken independence was a constant warning to the oligarchy of planters not to encroach too far upon the rights of free men. By the early 18th century, colonial legislatures held two significant powers: the right to vote on taxes and expenditures, and the right to initiate legislation rather than merely act on proposals of the governor. The legislatures used these rights to check the power of royal governors and to pass other measures to expand their power and influence. In time, the center of colonial administration shifted from London to the provincial capitals.

Reporter System Using LuxABCDE

Reporter System Using LuxABCDE La Rosa, S.L., Diep, D.B., Nes, I.F., et al. (2012) Construction and application of a luxABCDE reporter system for real-time monitoring of Enterococcus faecalis gene expression and growth. Applied and Environmental Microbiology, 78 (19): 7003-7011 Reporter genes are the genes encoding a protein which can be tracked and quantified by microscopy and various biochemical assays. They provide a non-invasive and sensitive method to monitor levels of gene expression, protein localization and to determine the transcriptional and translational regulators of a gene of interest. In this study, Rosa and co-workers have developed a bioluminescence based reporter system using luxABCDE to monitor the growth and gene expression of Enterococcus faecalis. E. faecalis is a nosocomial pathogen reported to be the cause of diseases such as endocarditis, urinary tract infection, bacteremia etc. (Schlievert et. al., 1998) and particularly infects individuals with compromised immune system or suffering from an underlying illness (Mundy et. al., 2000). Several studies have been undertaken to determine the factors involved in the virulence of the nosocomial strains of E. faecalis (Jett et. al., 1992; Chow et. al., 1993; Shankar et. al, 2001). These stud ies involve creating mutants for a possible virulence trait and then comparing them in animal models. There are several advantages of using reporter gene in lieu of traditional methods which usually involve harvesting the infected organ and sample preparation to estimate the level of infection by E. faecalis (Hanin et. al., 2010; Ike et. al., 1984). This traditional method, apart from being time consuming, also requires sacrifice of a large number of animals used in biological experiments. Using reporter genes, although not able to completely replace animal experimentation, will certainly reduce the number of animals required. Another advantage of using reporter genes is that it will enable real time monitoring of spread of infection by imaging the light emitted from the activation of the lux operon which was not possible with the other studies that have been undertaken. Use of lux operon also provides several advantages over other reporter gene based systems used in studies pertain ing to E. faecalis which employ green fluorescent protein (gfp) or firefly- luciferase enzyme (luc). Both gfp and luc require excitation by an external light source and luc also requires the addition of an extraneous substrate- both of these are not required in case of luxABCDE thus providing another advantage to the use of this reporter system. Rosa and co-workers constructed the bioluminescent reporter system by cloning the luxABCDE operon present in pPL2lux into pREG696 as shown in Figure 1. The native luxCDABE operon from Photohabdus luminescens shows poor expression in gram-positive bacteria (Qazi et. al., 2001). Therefore, the genes were rearranged into luxABCDE and translational signals were inserted in front of luxA, luxC, luxE to increase the expression of the lux operon. pPL2lux was originally designed for Listeria monocytogenes and thus carries a listeriophage integrase gene. This plasmid does not work in E. faecalis because of the absence of the required listeriophage sequence. Therefore, a highly expressed Listeria promoter Phelp (for highly expressed Listeria promoter) was cloned into pPL2lux just upstream of the luxA gene and this construct was named pPL2luxPhelp . Similarly, other promoters P32 and P16S were cloned upstream of luxA gene to form pPL2luxP32 and pPL2luxP16S respectively. P32 is another strong pr omoter and P16S is a synthetic E. faecalis 16sRNA promoter with a ribosome binding site (RBS) and an ATG codon fused at its end. These modified pPL2lux plasmids were digested with Xho1 and Not1 and the excised fragment (luxABCDE and the promoter) was cloned into the corresponding sites in pREG696 to form pSL101P32/16S/help. pREG696 has a spectinomycin resistance gene and a segregational stability cassette (axe- antitoxin and txe- toxin) from the multi-drug resistant plasmid pRUM of E. faecium (Grady and Hayes, 2003). This segregational stability cassette enables stable inheritance of the plasmid by killing of plasmid-free cells. All the DNA fragments inserted were sequenced before transferring them into E. faecalis MMH594 by electroporation. Figure 1 Schematic of the construction of pSL101 and its derivatives. (Figure reproduced from La Rosa et. al., 2012) The stability of pSL101P32, pSL101P16S, pSL101Phelp was tested by a doing a plasmid stability test. The plasmids were transformed into E. faecalis and overnight cultures were diluted 1,000-fold and grown in nonselective GM17 media. After every 24 hours a fresh culture was inoculated and this was continued for 7 days. At every inoculation, the culture was diluted and plated onto selective and nonselective plates and incubated at 37 °C overnight. Bioluminescence of the resulting colonies was measured and the number of colonies on both selective and nonselective plate was counted to score for resistance to spectinomycin. The stability of these  plasmids was compared to pIL252luxABCDEPhelp which was created by cloning the Phelp luxABCDE cassette into pIL252. This plasmid lacks the axe-txe stability system and is therefore lost after overnight culture with antibiotic selection (Figure 2a). On the other hand, there is no loss of pSL101P32 even after 7 days of culturing in non-selective media and only 30% loss in case of pSL101P16S and pSL101Phelp. This result shows that the segregational stability system is required for stable maintenance of the reporter plasmids in E. faecalis grown in nonselective media. The methodology adopted by the authors to do this experiment has two drawbacks- firstly, plating the culture onto nonselective and selective plates simultaneously may give erroneous results when there are more colonies on the selective plate than on the nonselective plate. To avoid this, the colonies on the nonselective plate obtained after incubation for 16 hours should be patched onto selective plates and then scored for antibiotic resistance phenotype. Secondly, nowhere in the manuscript do the authors mention doing the experiments in repeats. Performing an experiment in duplicates or triplicates is advantageous as it makes the data obtained more reliable. (Figure reproduced from La Rosa et. al., 2012) After confirming the stability of the plasmids in E. faecalis MMH594, overnight cultures were diluted and grown in GM17 medium and bioluminescence was measured to assess the correlation between light emission and cellular growth. To determine the relationship between CFU number and bioluminescent signal cultures in mid-exponential phase were diluted and viable cell count and bioluminescence was measured. Figure 2b shows that a linear relationship exists between number of CFU and the bioluminescent signal and thus pSL101 reporter system can be used to measure the real-time growth of bacteria. Also, to determine the relation between absorbance and bioluminescence growth of cultures with pSL101 derivatives was measured using a microplate reader by recording the absorbance at 620 nm for 7 hours after 15 min intervals. Bioluminescence of the culture was measured by quantifying the bioluminescent signal captured by an imaging system. The experiment was done in three independent repeats. It was found that during the exponential phase bioluminescence increased because of high metabolic activity of the cells and thus high availability of substrate- flavin mononucleotide (Bachmann et. al., 2007) required for luciferase enzyme (Figure 3). As the cells enter the stationary phase, a decline in the bioluminescence signal was observed corresponding to the decrease in the concentration of the substrate. Similar effect was observed for pSL101P32, pSL101P16S, pSL101Phelp. In case of the negative control, no bioluminescence was observed which confirms no background. Growth of different strains of E. faecalis was also monitored using the pSL101P16S system to determine its broad applicability. Four strains of E. faecalis differing in their origin (clinical isolate, probiotic strain, laboratory strain and commensal strain) were transformed with pSL101P16S and absorbance and bioluminescence was measured as above. As in the case of E. faecalis MMH594 a good correlation (R2 > 0.94) was observed between absorbance and bioluminescence thus proving that pSL101 system is not limited to a single strain of E. faecalis but can be used for other strains too. It is also commonly found in Therefore, pSL101P16S was used to monitor the growth of E. faecalis in these environments. To measure the growth in milk, Nestle NAN Infant Milk Formula was pre-heated to 37 °C and inoculated with PBS (phosphate-buffer saline) washed probiotic strain Symbioflor 1 and commensal strain E. faecalis 32. Both the strains were lux-tagged with pSL101P16S. Bioluminescence was measured as described above and number of CFU was measured at 2, 4, 6, 8 and 22 hours after inoculation by plating the cultures on GM17 media with spectinomycin. The experiment was done in duplicates. There was no background luminescence detected by milk and for both the strains, a linear correlation (R2= 0.95) between bioluminescence and viable cell count was observed (Figure 4). Similarly, growth of lux-tagged E. faecalis MMH594 and T2 was measured in urine by preheati ng the media at 37 °C and inoculation with PBS washed cells. Although a low level of luminescence was observed in urine, there was a significant increase in the luminescence corresponding to the growth of bacterial strains (Figure 5) as in the case of milk. This experiment shows that the pSL101 system can be used in other growth environments as long as the background luminescence due to media is low. G. mellonella larvae can be maintained at 37 °C thus permitting the study of host- pathogen interaction at the human physiological temperature. Also, they have a fairly advanced immune system comprising of phagocytic cells in the blood type fluid called hemolymph. To determine whether the luxABCDE cassette influences the virulence of E. faecalis MMH594, killing of G. mellonella larvae was monitored when infected by wild-type E. faecalis MMH594 and the strain tagged by pSL101P16S. This was done by injecting 10  µl of E. faecalis suspension (in 0.9% saline) into the body cavity of 10 larvae through the hindmost leg. As a control 10  µl of 0.9% saline was injected into another 10 larvae. These infected larvae were grown at 37 °C on 90 mm petri dishes and were examined every 2 hours. It was observed that the infection due to E. faecalis is accompanied by the melanisation of larvae whic h indicates towards the activation of prophenoloxidase (PO) responsible for biosynthesis of melanin and plays a role in the defence reactions against invading organisms (Sugumaran, 2001). From Figure 6a it can be confirmed that the virulence of lux-tagged strain of E. faecalis is similar to the wild type strain. Therefore, the lux-tagged strain was used to visualize the progress of infection in G. mellonella. Figure 6b shows the bioluminescent images of infected larvae captured till 48 hours after infection. The bioluminescent signal was detected immediately after infection and declined after 2 hours. After 4 hours post infection, a peak in the signal was observed and was constant till 24 hours after which all larvae were dead (Figure 6c). To confirm whether the bioluminescent signal corresponds to the growth of E. faecalis the infected larvae were sterilized by 70% ethanol and dissected and transferred into 0.9% saline solution. These samples were vortexed and homogenous mixture of insect and bacteria was serially diluted and plated on GM17 plated with spectinomycin. At each time period, 3 insects were dissected and number of CFU was counted. A drop in the number of CFU was observed 2 hours after infection but this was followed by increase in growth till 48 hours (Figure 6d). This result is in good agreement with the bioluminescent signal measured in the infected larvae and shows that the bioluminescent reporter can be used to monitor the progress of infection by E. faecalis in G. mellonella. (Figure reproduced from La Rosa et. al., 2012) The use of luxABCDE as a reporter for monitoring growth of E. faecalis in animal models has several advantages (as stated earlier). On the other hand, there are certain limitations of this system. Bioluminescence of luxABCDE relies on fatty acid synthesis and thus on the metabolic activity of the cells. Lower the metabolic activity lower will be the bioluminescent signal. Due to this, the pSL101 reporter system designed in this study may not be able to measure the growth of bacterial cells with low metabolic rate. This is especially disadvantageous in case of biofilm formation by E. faecalis in which the cells may have low metabolic activity. Also, the report does not describe the effect of pSL101 derivatives (with P32, P16S and Phelp) on the growth of E. faecalis. Although, it is mentioned that the growth of E. faecalis MMH594 strain with pSL101 is similar to the strain with pSL101P32, pSL101P16S, pSL101Phelp (data not shown) it is necessary to investigate whether the pSL101 and its derivatives present any metabolic load on the E. faecalis strain. The study shows that the pSL101 derivatives are stably maintained in E. faecalis but the plasmid stability test was done only for duration of 7 days and it is possible that the plasmids may be rapidly lost after this time period. This will prevent the use of the pSL101 reporter system to monitor growth of E. faecalis in other animal models, such as mouse, where long-time monitoring is required. Therefore, it is necessary to assess the stability of these plasmids in E. faecalis over a long time. Additionally, it may be difficult to observe any bioluminescent signal from deep tissues of animal models if the signal is weak, as in the case of low number of E. faecalis cells. In conclusion, a simple and robust reporter system using luxABCDE has been developed to monitor the growth of E. faecalis in animal models. Despite the limitations, this method is non-invasive and will significantly reduce the burden on experimental animals. It can be used to investigate the various genes involved in the virulence of E. faecalis facilitating better understanding of pathogenicity of E. faecalis. Dwight’s Early Life Dwight D. Eisenhower was the third son of David and Ida Stover Eisenhower. He was born in 1890 in Denison, Texas, and named David Dwight Eisenhower, although he was known as Dwight David by many. In 1891, the family moved to Abilene, Kansas, where Eisenhower was brought up. He was the third of seven sons. He and his older brothers were all called â€Å"Ike† by their family, Eisenhower was known as â€Å"Little Ike†. In his high school years, he was known to excel in sports due to his active nature. After he graduated, Eisenhower wanted to attend college, but his family could not afford the tuition. Dwight and his brother planned to switch off between work and college every year in order to pay for each other’s tuition and allow them to both complete their education. In 1910, Eisenhower found that he could get a free college education at United States Military Academy at West Point, New York. The prerequisite for obtaining such involved passing a difficult exam. W hile Eisenhower had no original plans to be a soldier, he still prepared well for the competitive West Point entrance exam and won an appointment to the school in 1911. The Coming of a Commander in Chief Unknown to him at the time, Eisenhower would later lead many military forces though the course of both world wars, winning decisive victories and helping push America forward even before his own presidency. When the United States entered World War I in 1917, Eisenhower was promoted in the army and assigned to training duty for new cadets. He desperately wanted to see action during the war, and applied for an overseas assignment. His own skill would prevent him from participating in battle during that war. Higher officers saw the ability that he had as an organizer and trainer, and put him in command of Camp Colt at Gettysburg, Pennsylvania, instead of granting his overseas request. One of the army’s first tank corps was being formed there, and Eisenhower trained the fighting unit. In the October of 1918 he finally got orders to take his units overseas, to France, but the war ended before he could leave America. Although disappointed at having missed combat, Eisenhower was reco gnized by his superiors for his efforts during the war. He was awarded the Distinguished Service Medal following the submission of German forces.
